New CLINPROT magnetic beads

The new Glyco Kit MB-LAC WGA (Wheat Germ Agglutinin) complements the glyco bead product line. While the existing Glyco Kits MB-LAC ConA and MB-CovAC Boronic are suitable for capturing of glycosylated proteins and peptides in a more general manner, the MB-LAC WGA captures respective molecules with a more specific binding motif:

The receptor sugar for WGA is N-acetylglucosamine preferentially binding to dimers and trimers of this sugar. WGA can also bind oligosaccharides containing terminal sialic acids or chitobiose.

ConA (Concanavalin A) binds to most N-glycosylated proteins, due to the conserved penta-core structure of glycoproteins containing mannosyl and glucosyl residues with free hydroxyl groups at positions C3, C4 and C6.

Boronic acid forms covalent bonds to molecules containing cis-diol groups, like mannose, galactose or glucose and captures therefore in addition to N-glycosylated also O-glycosylated and C-glycosylated proteins carrying respective groups.

Bruker Daltonics Glyco Kits allow direct capturing of glycosylated peptides and proteins from human serum or other complex protein sources and the subsequent characterization as well as identification by MALDI-TOF and MALDI TOF/TOF analysis.

Combining the respective specific glyco-capturing beads, tryptic digest and MB-HIC C8 purification facilitates identification of serum glycoproteins contributing to the fraction making up 1% of serum proteins. Each bead type presents its own individual binding profile that partially also overlaps with the profiles of the two others functionalities.
