MALDI-TOF Mass Spectrometry based Genotyping Platform

The GENOLINK platform is a suite of tools for semi-automated single nucleotide polymorphism genotyping.
Overview
PCR products are processed with the genoSNIP assay, which incorporates a non-nucleotide UV-cleavable linkage into a biotinylated primer. Using this primer, a primer extension reaction will generate allele-specific products which subsequently are purified with the genostrepTM purification kit. After UV-cleavage the extension products are ready to be measured with high accuracy in the MALDI-TOF mass spectrometer. Alternatively, natural primers can be used in combination with genopureTM purification system.

Sample preparation

The genostrep kit is designed for the purification of single-stranded DNA-molecules after primer extension reactions carried out with biotinylated primers containing a UV-cleavable linker. The purification procedure is based on the strong streptavidin-biotin binding and removes salts, nucleotides and other undesirable components from the extension mixture. After UV-cleavage, the purified products are ready for measurement in a MALDI-TOF mass spectrometer.
- Alternatively, genopure dsTM and genopure oligo™ kits for ultimate quality purification of PCR products and single stranded DNA with Magnetic Beads
The genopure ds kit is designed for the purification of double stranded DNA like PCR-products or restriction enzyme fragments from nucleotides, primers, salts and other undesirable components based on binding onto the surface of magnetic beads. PCR-products of 40 up to several 1000 nucleotides are purified rapidly and recovered in an elution buffer.
The genopure oligo kit is qualified for purification of single stranded DNA molecules like oligonucleotides, primer extension products and sequencing reaction products from contaminating reaction components.
After the purification the DNA-molecules are ready for measurement in a MALDI-TOF mass spectrometer.

By genotools the existing Bruker Daltonics proteomics bioinformatics product line is extended to a functional genomics solution. This offers the unique advantage of the combined usage of one instrument platform for proteomics and genomics applications.
