Bruker ClinProt solution. They offer a wide range of functionalities leading to an enormous range of possible applications. Bruker Daltonics ClinProt Kits contain all buffers required and are dedicated for the subsequent direct characterization as well as identification by MALDI-TOF and MALDI TOF/TOF analysis. Different analysis strategies are possible as well.
General Profiling of Body Fluids
ClinProt Profiling Kits are especially designed for sophisticated biomarker profiling studies. Combining the three different appropriate and dedicated bead functionalities (hydrophobic or cation exchange interaction or affinity to immobilized copper ions) allow systematical and intelligent biomarker discovery workflows within a huge mass range from 1000 Da up to 70.000 Da.
Body Fluid Profiling with Focus on Glycoproteins
Glycoprotein profiling of body fluids e.g. serum or cerebrospinal fluid (CSF) is supported by the surpassing variety of Glyco Beads since each bead type is directed to a different receptor sugar. Each bead type presents its own individual binding profile that partially also overlaps with the profiles of the others functionalities. Glycobeads are also successfully used for specific capturing and purification of glycosylated proteins and peptides from other complex protein sources.
Enrichment and Purification of Peptides / Proteins with Affinity to Metal
The affinity of peptides and proteins to metal ions can be used for their purification, selective capturing or enrichment. Prominent examples are the enrichment of His-tagged proteins based on their affinity to Ni2+ and/or Co2+ or the capturing of phosphopeptides based on their affinity to Fe3+, Al3+ or Ga3+. After capturing, the metal binding proteins are directly analyzed e.g. by MS and MS/MS.
Specific Capturing based on protein-protein or antigen-antibody interactions
Complexity of proteomic samples and the growing interest in exploring protein-protein interactions require techniques for specific and simple enrichment of proteins or a protein’s interaction partners. Proteins can be extracted very specifically if its respective antigen is bound to magnetic beads whereas protein-protein interactions can be studied by immobilizing a protein and subsequent capturing of ligands and interaction partners combined with MS detection and identification. Subsequent analysis is possible without any further purification steps.
Purification of peptides and proteins
Peptides and proteins often require desalting and the removal of accompanying small molecules to show clear mass spectra with satisfying signal intensities. Magnetic beads with hydrophobic surface are shown to be an ideal tool for such a purification approach.
